چکیده

The aim of the present study is to investigate the effect and mechanism of recombinant human acidic fibroblast growth factor (rhaFGF) on skin wound healing of diabetic rats. SD rats were randomly divided into three groups: control, diabetes and rhaFGF groups. Skin wound tissues of rats in the three groups were respectively arranged on days 3 and 7 after establishment of skin wound of diabetic rats. Immunohistochemistry analyses were performed to evaluate the effect of rhaFGF on expression levels of PCNA and CD34 in diabetic rats. Real-time PCR and Western blot assays were used to examine the expression levels of Bcl-2, Bax, TGF-β, CXCL-1, CXCL-2, p-ERK1/2 and p-STAT3 in wound tissues. The results showed that rhaFGF could significantly increased expression levels of PCNA, CD34, TGF-β, CXCL-1, CXCL-2, p-ERK1/2, p-STAT3 and the ratio of Bcl-2/Bax in wound tissues. The preliminary mechanism that rhaFGF accelerates wound healing of diabetic rats is possibly associated with the promotion of fibroblasts proliferation and angiogenesis through ERK1/2 and STAT3 pathways.

Aloe vera oral administration accelerates acute radiation-delayed wound healing by stimulating transforming growth factor-β and fibroblast growth factor production.

BACKGROUND Delayed wound healing is a significant clinical problem in patients who have had previous irradiation. This study investigated the effectiveness of Aloe vera (Av) on acute radiation-delayed wound healing. METHODS The effect of Av was studied in radiation-exposed rats compared with radiation-only and control rats. Skin wounds were excised on the back of rats after 3 days of local ra...

Novel Anti-Microbial Peptide SR-0379 Accelerates Wound Healing via the PI3 Kinase/Akt/mTOR Pathway

We developed a novel cationic antimicrobial peptide, AG30/5C, which demonstrates angiogenic properties similar to those of LL-37 or PR39. However, improvement of its stability and cost efficacy are required for clinical application. Therefore, we examined the metabolites of AG30/5C, which provided the further optimized compound, SR-0379. SR-0379 enhanced the proliferation of human dermal fibrob...

Impaired healing of gastric lesions in streptozotocin-induced diabetic rats: effect of basic fibroblast growth factor.

We examined the influence of diabetes on the healing of HCI-induced gastric lesions and the healing promoting effect of basic fibroblast growth factor (bFGF) on these lesions under diabetic conditions induced in rats by streptozotocin (70 mg/kg, i.p.). The experiments were performed using 2-wk streptozocin-diabetic rats with blood glucose levels of >300 mg/dl. After 18 hr fasting, these animals...

Notoginsenoside Ft1 Promotes Fibroblast Proliferation via PI3K/Akt/mTOR Signaling Pathway and Benefits Wound Healing in Genetically Diabetic Mice.

Wound healing requires the essential participation of fibroblasts, which is impaired in diabetic foot ulcers (DFU). Notoginsenoside Ft1 (Ft1), a saponin from Panax notoginseng, can enhance platelet aggregation by activating signaling network mediated through P2Y12 and induce proliferation, migration, and tube formation in cultured human umbilical vein endothelial cells. However, whether it can ...

بررسی اثر فاکتور رشد انسولینی در روند ترمیم زخم های حاد پوستی در رت های دیابتی شده به وسیله استرپتوزوتوسین

Abstract Background and purpose: Delayed wound healing is a common complication of diabetes mellitus. However, less is known about the cause of this pathology. Types of skin cells, extracellular matrix and variety of growth factors are involved in wound healing. The use of recombinant growth factors in researches and production of skin substitutes are still a challenge. Much research has bee...
